目的 观察重组β-防御素-2对呼吸道绿脓杆菌感染大鼠急性肺损伤(ALI)的保护作用。 方法10只清洁级雄性成年SD大鼠,随机分为防御素组和对照组,每组5只。防御素组大鼠暴露声 门后,气管内滴注5×107PFU/ml重组腺病毒(含有β-防御素-2编码基因)50μl,对照组给予等量对照腺 病毒(不含β-防御素-2编码基因)。48 h后两组气管内滴注6×108CFU/ml绿脓杆菌ATCC27853 200μl, 制备绿脓杆菌感染致ALI模型。气管内滴注绿脓杆菌24 h后处死大鼠,采集肺泡灌洗液,进行绿脓杆 菌菌落数和白细胞计数;观察肺组织病理学变化,并测定肺组织细胞间粘附分子-1(ICAM-1)表达水 平。结果与对照组比较,防御素组BALF中绿脓杆菌菌落数、白细胞计数、肺组织ICAM-1表达水平 及肺病理组织学评分降低(P<0.05)。结论重组β-防御素-2对呼吸道绿脓杆菌感染大鼠ALI有一 定的保护作用,可能与其杀菌作用和下调肺组织ICAM-1的表达有关。
Objective To observe the protective effect of recombinant β-defensin-2 on acute lung injury (ALI) induced by respiratory tract Pseudomonas aeruginosa in rats. Methods Ten male adult SD rats were randomly divided into defensin group and control group, with 5 rats in each group. After the defensin group was exposed to the glottis, 50μl of 5 × 107PFU / ml recombinant adenovirus (containing the β-defensin-2 encoding gene) was instilled into the trachea, and the control group was given the same amount of control adenovirus (without β-defensin -2 encoding gene). After 48 h, two groups of tracheal instillation of 6 × 108CFU / ml Pseudomonas aeruginosa ATCC27853 200μl prepared Pseudomonas aeruginosa infection caused by ALI model. Intratracheal instillation of Pseudomonas aeruginosa 24 h after the rats were sacrificed, the alveolar lavage fluid was collected for Pseudomonas aeruginosa colonies and white blood cell count; observe the pathological changes of lung tissue, and determination of lung tissue intercellular adhesion molecule-1 (ICAM -1) expression level. Results Compared with the control group, the number of P. aeruginosa BALB / c mice, leukocyte count, expression of ICAM-1 in lung tissue and histopathological score of lung in defensin group decreased (P <0.05). Conclusion Recombinant β-defensin-2 can protect ALI induced by Pseudomonas aeruginosa in rats, which may be related to its bactericidal effect and the down-regulation of ICAM-1 expression in lung tissue.
