病毒感染的快速诊断对治疗很有意义。据报告,用抗水痘、带状疱疹病毒(VZV)单克隆抗体荧光抗体直接法,能检出局部病变中的VZV特异性糖蛋白抗原。作者用直接法对带状疱诊、水痘和单纯疱疹的诊断和鉴别等进行探讨,现报告如下。材料和方法:带状疱疹60例,水痘16例,单纯疱疹19例。采集病变处水疱底部的细胞成分、痂皮、角膜擦拭物和脑脊液等。另采集46例带状疱疹血清,进行血清补体结合反应。方法系将采集的材料涂于载玻片上,痂皮在生理盐水中浸泡30分钟后涂片,或立即冰冻后用恒冷切片机切片后贴附于玻片上。使其与VZV特异的糖蛋白导硫氰酸荧光素(FITC) 标记的鼠单克隆抗体发生反应,用荧光显微镜确定抗原,全过程只需1小时左右。结果:60例带状疱疹的水疱底成分抗原阳性59例,3例有眼部病变的角膜擦拭物均阳性,脑脊液细胞检查带状疱疹1例阴性。16例水痘均阳性。对15例
Rapid diagnosis of viral infection is of great value to the treatment. It has been reported that the VZV-specific glycoprotein antigen in local lesions can be detected by the direct method using the anti-chickenpox and zoster virus (VZV) monoclonal antibody fluorescent antibody. The authors use the direct method of diagnosis of the blister blisters diagnosis, varicella and herpes simplex are discussed, are as follows. Materials and Methods: 60 cases of shingles, 16 cases of chickenpox, 19 cases of herpes simplex. The lesion at the bottom of the blisters collected cellular components, crusts, corneal wipes and cerebrospinal fluid. In addition, 46 cases of herpes zoster sera were collected for serum complement fixation. Method is to collect the collected material on glass slides, scabs in saline soaked for 30 minutes after the smear, or immediately after freezing with a constant temperature cold section slicer attached to the slide. It reacted with mouse monoclonal antibody labeled with fluorescein thiocyanate fluorescein (FITC), a VZV-specific glycoprotein. Fluorescence microscopy was used to determine the antigen. The whole process took about 1 hour. Results: In the 60 cases of herpes zoster, 59 cases were positive for blister component antigen, 3 cases were positive for corneal swab with ocular lesions, and 1 case was negative for cerebrospinal fluid cells examination. 16 cases of chicken pox were positive.
