目的分析大连地区鼠携带汉坦病毒(HV)情况及基因特征。方法用间接免疫荧光方法检测大连地区2012年采集的鼠肺,用特异性引物扩增阳性标本中M基因片段,测序后进行生物信息学分析,构建系统进化树,以明确其型别和亚型。结果检测鼠肺188份,其中黑线姬鼠3份,褐家鼠185份,检出HV抗原阳性6份,阳性率为3.19%。所携带的病毒均为汉城型病毒(SEOV),S3亚型。结论大连地区鼠携带HV呈地区聚集性分布,并且可能存在除S3外的其他亚型,后续应系统、连续地开展HV的分子流行病学研究以进一步明确其亚型及分布。
Objective To analyze the status of hantavirus (HV) and its genetic characteristics in rats in Dalian area. Methods Indirect immunofluorescence assay was used to detect the murine lungs collected in 2012 in Dalian. The M gene fragments of positive specimens were amplified by specific primers. Bioinformatics analysis was performed after sequencing, and phylogenetic tree was constructed to confirm the type and subtype . Results A total of 188 rat lungs were examined, including 3 Apodemus agrarius and 185 Rattus norvegicus. Six positive HV antigens were detected, the positive rate was 3.19%. The viruses carried are all Seoul virus (SEOV), S3 subtype. CONCLUSION: The regional HV distribution in rats in Dalian is clustered and there may exist other subtypes except S3. The molecular epidemiology of HV should be continuously and systematically studied to further clarify its subtypes and distribution.
