目的:建立胡颓子叶中银锻苷含量测定的方法。方法:采用80%甲醇提取样品;HPLC法测定,色谱条件为:色谱柱为Agilent C_(18)(250 mm×4.6 mm,5μm),流动相:乙腈-0.1%磷酸水溶液,梯度洗脱,柱温:30℃,流速:1.0 ml·min~(-1),检测波长:315 nm,进样量:20μl。结果:银锻苷在0.01~0.20 mg·ml~(-1)范围内与峰面积积分值呈良好的线性关系(r=0.999 9),平均回收率为99.14%,RSD为1.33%(n=6)。结论:该方法简便、快速、准确、重复性好,可用于胡颓子叶的含量测定。
Objective: To establish a method for determination of silver forgivain in leaves of Elaeagnus. Methods: The samples were extracted with 80% methanol and determined by HPLC. The chromatographic conditions were as follows: Agilent C 18 (250 mm × 4.6 mm, 5 μm), mobile phase of acetonitrile-0.1% phosphoric acid Temperature: 30 ℃, flow rate: 1.0 ml · min ~ (-1), detection wavelength: 315 nm, injection volume: 20μl. Results: The results showed that there was a good linearity (r = 0.999 9) in the range of 0.01 ~ 0.20 mg · ml ~ (-1) with an average recovery of 99.14% and a RSD of 1.33% (n = 6). Conclusion: The method is simple, rapid, accurate and reproducible. It can be used to determine the content of leaves in Elaeagnus.
