目的:评价超临界分散法制备的5-氨基水杨酸(5-ASA)结肠定位纳米粒对小鼠结肠炎的治疗作用。方法:将24只6周龄BABL/c小鼠随机分为空白对照组,结肠炎模型组,5-ASA治疗组,5-ASA纳米粒治疗组。三硝基苯磺酸(TNBS)造模后的第2-7 d分别给予0.9%氯化钠,5-ASA和5-ASA纳米粒灌胃,于第8 d处死所有小鼠,评估小鼠的疾病活动指数(DAI),组织病理形态以及结肠组织学损伤情况;检测结肠组织髓过氧化物酶(MPO)活性。结果:5-ASA和5-ASA纳米粒均能明显减轻结肠重量,缩小溃疡面积,降低MPO活性。超临界分散法制备的5-ASA纳米粒在DAI,减轻局部的炎症程度(MPO水平)方面,其疗效与5-ASA组(50 mg/Kg)相当。结论:超临界分散法制备的5-ASA纳米粒对TNBS诱导小鼠的结肠炎有明显的治疗作用。
Objective: To evaluate the therapeutic effect of 5-aminosalicylic acid (5-ASA) colloidal nanoparticles prepared by supercritical dispersion on colitis in mice. Methods: 24 6-week-old BABL / c mice were randomly divided into blank control group, colitis model group, 5-ASA treatment group and 5-ASA nanoparticles treatment group. The mice were sacrificed on the 8th day after intraperitoneal injection of 0.9% sodium chloride, 5-ASA and 5-ASA nanoparticles on the 2-7th day after trinitrobenzene sulfonic acid (TNBS) (DAI), histopathology and histopathological damage of colonic tissue. The activity of myeloperoxidase (MPO) in colonic tissue was detected. Results: Both 5-ASA and 5-ASA nanoparticles could obviously reduce the weight of colon, reduce the ulcer area and decrease the activity of MPO. The 5-ASA nanoparticles prepared by the supercritical dispersion method have the same curative effect as the 5-ASA group (50 mg / Kg) in terms of DAI and alleviation of local inflammation (MPO level). CONCLUSION: 5-ASA nanoparticles prepared by supercritical dispersion have a significant therapeutic effect on TNBS-induced colitis in mice.
