建立测定血清中胆固醇酯转运蛋白(CETP)活性的方法.方法 用14C标记的胆固醇酯合成高密度脂蛋白(HDL)样颗粒作为反应基质,检测了45例健康人、52例冠心病(CHD)患者血清CETP活性.结果 该方法线性范围为0-31.8%;高(26.4%)、低(6.83%)活性批内变异系数(CV)分别为6.1%、7.6%,批间CV分别为11.7%、12.1%,52例CHD患者血清CETP活性(x±s)为17.7%±5.3%,明显高于健康对照组(12.8%±4.1%),(P<0.01).结论 该法简便快速、精密度高,为测定CETP活性提供了一种较好的方法.“,” Objective To establish a new method for determination of cholesteryl ester transfer protein CETP activity in serum. Method The assay was based on 14C cholesteryl ester-labeled discoidal bilayer particles DBP instead of HDL as donor and purified LDL as the acceptor. The serum CETP activity of 45 healthy subjects and 52 patients with coronary heart disease CHD was examined. Results The assay showed linearity from 0 to 31.8% with high 26.4% and low activity 6.83%. The within-run CV were 6.1% and 7.6% respectively and the between-run CV 11.7% and 12.1%. CETP activity measured in 52 CHD patients was x±s=17.7±5.3% which was significantly higher than that of the control group x±s=12.8±4.1% P<0.01. Conclusion It is an easy rapid and precise method offering a better way for determining CETP activity.
