目的本实验通过维甲酸对乳腺癌细胞的钠碘转运体(NIS)基因及蛋白表达和摄碘功能的影响,探讨乳腺癌细胞NIS的表达特点和调控因素。方法我们采用乳腺癌细胞MDA-MB453进行培养,分别给予不同浓度的维甲酸(RA)进行刺激72h。采用RT-PCR方法检测细胞中NIS的mRNA表达;用WesternBloot方法检测NIS蛋白水平;采用125I测定乳腺癌细胞摄碘功能的变化。结果乳腺癌细胞MDA-MB453在经过RA刺激后,细胞中NIS的mRNA表达比对照组有明显增加,并且与RA浓度成正比。细胞中NIS的蛋白表达比对照组也有明显增加,并且与RA浓度相关。经过RA刺激的乳腺癌细胞摄碘能力明显增加。结论维甲酸对乳腺癌细胞MDA-MB453的NIS基因和蛋白表达均有促进作用,而且促进乳腺癌细胞的摄碘,这提示维甲酸可能通过调节乳腺癌NIS基因和蛋白的表达,从而影响乳腺癌细胞对放射性碘治疗的敏感性。
OBJECTIVE: To investigate the effect of retinoic acid on the expression of sodium iodide transporter (NIS) and the function of iodine uptake in breast cancer cells, and to explore the expression characteristics and regulatory factors of NIS in breast cancer cells. Methods We cultured breast cancer cells MDA-MB453 and were given different concentrations of retinoic acid (RA) for 72h. The mRNA expression of NIS was detected by RT-PCR. The protein level of NIS was detected by Western Blot. The iodine uptake by breast cancer cells was determined by 125I. Results The mRNA expression of NIS in breast cancer cells MDA-MB453 after RA stimulation increased significantly compared with the control group and was directly proportional to the concentration of RA. The protein expression of NIS in cells was also significantly increased compared with the control group, and correlated with the RA concentration. RA-stimulated breast cancer cells significantly increased iodine uptake capacity. Conclusion Retinoic acid can promote the expression of NIS gene and protein in breast cancer cell line MDA-MB453 and promote the uptake of iodine by breast cancer cells. This suggests that retinoic acid may affect the expression of NIS gene and protein in breast cancer and thus affect breast cancer Cell sensitivity to radioactive iodine therapy.
