目的分析T-box转录因子3(T-box transcription factor 3,TBX3)与小鼠双微体基因(murine double mimute 2,MDM2)在喉鳞状细胞癌(简称喉鳞癌)组织中的表达水平及其与临床病理特征的相互关系,并探讨其在喉鳞癌发生发展过程中可能的作用。方法回顾性分析55例喉鳞癌患者临床病理资料,分别用实时荧光定量PCR(real-time quantitative PCR,RQ-PCR)和免疫组化法检测喉鳞癌组织及相应的癌旁组织中TBX3与MDM2的mRNA和蛋白表达水平。结果 TBX3与MDM2mRNA在喉鳞癌组织中均呈过表达状态(t分别为13.226,13.504,P均为0.000),TBX3的表达与肿瘤的分化水平相关(χ2=9.728,P=0.002),MDM2的表达与肿瘤的临床分期、淋巴结转移相关(χ2分别为7.834、13.257,P分别为0.005、0.000)。TBX3蛋白定位肿瘤细胞的细胞核,MDM2蛋白主要定位于细胞核与细胞浆,二者在蛋白水平上的表达结果与RQ-PCR一致。TBX3与MDM2 mRNA在喉鳞癌组织中的表达水平呈正相关(r=0.479,P=0.000)。结论 TBX3与MDM2基因均在喉鳞癌中过表达且二者之间的表达水平呈正相关,联合检测喉鳞癌中的TBX3和MDM2可以作为喉鳞癌早期诊断和预后判断的重要指标之一。
Objective To analyze the expression of T-box transcription factor 3 (TBX3) and murine double mimute 2 (MDM2) in laryngeal squamous cell carcinoma (LSCC) And its relationship with clinicopathological features, and explore its possible role in the occurrence and development of laryngeal squamous cell carcinoma. Methods The clinicopathological data of 55 patients with laryngeal squamous cell carcinoma were retrospectively analyzed. The expression of TBX3 and its adjacent tissues were detected by real-time quantitative PCR (RQ-PCR) and immunohistochemistry MDM2 mRNA and protein expression levels. Results TBX3 and MDM2 mRNA were overexpressed in laryngeal squamous cell carcinoma (t = 13.226,13.504, P = 0.000). The expression of TBX3 was correlated with tumor differentiation (χ2 = 9.728, P = 0.002) Expression and clinical stage of the tumor, lymph node metastasis (χ2 were 7.834,13.257, P respectively 0.005,0.000). TBX3 protein localized in the nucleus of tumor cells. MDM2 protein mainly localized in the nucleus and cytoplasm. The expression of both proteins at the protein level was consistent with that of RQ-PCR. The expression of TBX3 and MDM2 mRNA in laryngeal squamous cell carcinoma was positively correlated (r = 0.479, P = 0.000). Conclusion Both TBX3 and MDM2 genes are overexpressed in laryngeal squamous cell carcinoma and there is a positive correlation between them. Combined detection of TBX3 and MDM2 in laryngeal squamous cell carcinoma can be used as one of the important indicators for early diagnosis and prognosis of laryngeal squamous cell carcinoma.
