目的:探讨清肺口服液含药血清对腺病毒3I、7b感染的人胚肺成纤维细胞之肿瘤坏死因子-α(TNF-α)mRNA基因表达的影响。方法:以3I、7b型腺病毒分别攻击体外培养的人胚肺成纤维细胞,制备清肺口服液含药血清,作用于该细胞,另设正常细胞组,病毒对照组,利巴韦林组,采用原位杂交法检测不同组细胞TNF-α的mRNA基因表达,并进行比较。结果:病毒对照组较正常细胞组之TNF-α mRNA基因表达明显增强(P<0.01),含药血清组可显著降低3I、7b型腺病毒攻击后细胞TNF-α的mRNA表达(P<0.01)。结论:①腺病毒感染可使人胚肺成纤维细胞TNF-α mRNA表达增高;②清肺口服液可下调TNF-α的mRNA表达,这可能是其抗病毒作用的机制之一。
Objective: To investigate the effect of Qingfei oral drug serum on the expression of tumor necrosis factor-α (TNF-α) mRNA in human embryonic lung fibroblasts infected by adenovirus 3I and 7b. METHODS: Adenovirus 3I and 7b were used to challenge human embryonic lung fibroblasts cultured in vitro to prepare Qingfei oral drug-containing serum. The cells were exposed to normal cells, virus control group and ribavirin group. The mRNA expression of TNF-α in different groups of cells was detected by in situ hybridization and compared. Results: The expression of TNF-α mRNA in the virus control group was significantly higher than that in the normal cell group (P<0.01), and the TNF-α mRNA expression was significantly reduced in the drug-containing serum group after 3I and 7b adenovirus challenge (P<0.01). ). Conclusion: 1 Adenovirus infection can increase the expression of TNF-α mRNA in human embryonic lung fibroblasts; 2 Qingfei Oral Liquid can down-regulate the mRNA expression of TNF-α, which may be one of the mechanisms of its antiviral effect.
