为了探索生长素酰胺合成酶(GH3)基因家族在桃(Prunus persica L.Batsch)果实生长发育过程中的作用,对桃基因组中的GH3基因家族进行生物信息学分析,对它们在溶质型和硬质型油桃果实发育成熟阶段的表达水平进行q RT-PCR检测。检测结果表明:桃GH3基因家族有8个成员,分别为Pp GH3-1~Pp GH3-8,聚类分析表明其分为GroupⅠ和GroupⅡ两类。实时荧光定量PCR分析表明GroupⅡ型基因(Pp GH3-1、Pp GH3-2、Pp GH3-3、Pp GH3-4和Pp GH3-5)在溶质型油桃中的表达水平高于硬质型油桃,其中Pp GH3-3和Pp GH3-4最为明显。经NAA处理后的硬质型油桃果实Pp GH3-1、Pp GH3-3、Pp GH3-4和Pp GH3-7相比对照上调表达,其中Pp GH3-3上调表达最为明显。试验结果表明生长素在桃果实成熟软化过程扮演着重要的角色。
In order to explore the role of the GH3 gene family in the fruit growth and development of Prunus persica L. Batsch, the GH3 gene family in the peach genome was bioinformatically analyzed, The expression level of mature nectarine fruit was detected by q RT-PCR. The results showed that there were 8 members of peach GH3 gene family, which were Pp GH3-1 ~ Pp GH3-8, respectively. Cluster analysis indicated that they were divided into GroupⅠ and GroupⅡ. Real-time quantitative PCR analysis showed that the expression level of Group Ⅱ genes (Pp GH3-1, Pp GH3-2, Pp GH3-3, Pp GH3-4 and Pp GH3-5) in solute nectarines was higher than that of hard oil Peach, of which Pp GH3-3 and Pp GH3-4 most obvious. Compared with the control, the expression of Pp GH3-3 was up-regulated in NAA treated Pp GH3-1, Pp GH3-3, Pp GH3-4 and Pp GH3-7. The results show that auxin plays an important role in the ripening and softening of peach fruits.
