[目的]观察中国汉族和朝鲜族8个Y-STR基因座遗传多态性在法医学个人识别和亲权鉴定中的适用性.[方法]利用PCR扩增、变性聚丙烯酰胺凝胶电泳和银染方法对各200名中国汉族和朝鲜族人群的DYS441~DYS448 8个Y-STR基因座单倍型进行检测和分析.[结果]在汉族群体中分别检测出7,5,6,7,4,10,10和6个等位基因,基因多样性分布为0.540 1~0.819 0,8个基因座联合单倍型为189种,基因多样性为0.999 2;在朝鲜族群体中分别检测出8,6,6,5,6,10,9和6个等位基因,基因多样性分布为0.513 5~0.799 6,8个基因座联合单倍型为174种,基因多样性为0.997 9.[结论]8个YSTR基因座在2个群体中均具有较高的遗传多样性,是较理想的遗传标记系统,对中国汉族和朝鲜族的个人识别及亲权鉴定等具有较高的应用性.
[Objective] To observe the applicability of 8 Y-STR loci genetic polymorphisms in Chinese foreigners and Koreans in forensic personal identification and paternity test. [Methods] PCR amplification, denaturing polyacrylamide gel electrophoresis and silver staining Methods The haplotypes of 8 Y-STR loci of DYS441 ~ DYS448 in 200 Chinese Han and Korean ethnic groups were detected and analyzed. [Results] In Han population, 7, 5, 6, 7, 10, 10 and 6 alleles. The distribution of gene diversity was 0.5401-0.8190. There were 189 haplotype haplotypes in 8 loci and 0.999 2 genotypes. Among the Korean ethnic groups, 8, 6, 6, 5, 6, 10, 9, and 6 alleles. The distribution of genetic diversity was 0.513 5-0.799 6, and 174 haplotypes were found in 8 loci with a genetic diversity of 0.997 9. [Conclusions ] The 8 YSTR loci are highly genetic diversity in 2 populations, which is an ideal genetic marker system and has high applicability to Chinese Han and Korean nationality identification and paternity test.
