目的 :观察小鼠烧伤前后肠道免疫分泌的变化和大黄汤治疗对正常小鼠和烧伤小鼠肠道免疫分泌的促进作用 ;探讨大黄保护肠粘膜屏障的作用机理。方法 :实验用BALB/c小鼠。分为 4组 :1组为对照组 (n =8) ;2组为正常小鼠饲喂 10 %大黄液 12ml/kg ,每 6h 1次 (n =11) ;3组为 30 %的全厚背部皮肤烧伤 ,不治疗 (n =7) ;4组为全厚皮肤烧伤 ,同 3组 ,大黄治疗同 2组 (n =9)。 2 4h后处死动物 ,灌洗肠道 ,收集肠液透析离心后作IgA检测和总蛋白 ,补体C3,高密度脂蛋白 (HDL)测定。结果 :1、2、3和 4组小鼠肠液中的IgA(mg/L)分别为 93 5± 13 7、15 0 8± 4 4 6、4 3 4± 2 1 5及 5 9 8± 19 3,说明在烧伤后肠液中IgA明显减少 (P <0 0 5 )。而应用大黄汤治疗后 ,无论正常小鼠和烧伤小鼠的肠道IgA均增加 (P <0 0 5 ) ,其他免疫相关物质如总蛋白、补体C3和HDL大黄治疗后也有显著增加。结论 :烧伤后小鼠肠液中的IgA明显减少。而大黄无论对正常小鼠和烧伤小鼠均具有明显的促进肠道免疫分泌的效果。这一作用是大黄的粘膜屏障保护作用的重要机制之一。
Objective : To observe the change of intestinal immune secretion before and after burn and the effect of Dahuang Decoction on intestinal immune secretion in normal mice and burned mice. To explore the protective mechanism of rhubarb protecting intestinal mucosal barrier. Methods: BALB/c mice were used for the experiment. Divided into 4 groups: Group 1 was control group (n = 8); Group 2 was normal mice fed with 10% rhubarb fluid 12ml/kg, every 6h (n = 11); 3 groups were 30% full thickness The back skin was burned and not treated (n = 7). The 4 groups were full-thickness skin burns. In the same group, the rhubarb treatment was in the same group (n = 9). After 24 hours, the animals were sacrificed, intestinal lavage was performed, intestinal fluid was collected and subjected to dialysis centrifugation for IgA detection and total protein, complement C3, and high-density lipoprotein (HDL) determination. RESULTS: IgA (mg/L) in intestinal fluid of mice of groups 1, 2, 3, and 4 were 93 5 ± 13 7, 15 0 8 ± 4 4 6, 4 3 4 ± 2 1 5, and 5 9 8 ± 19, respectively. 3, IgA in the intestinal fluid after burn was significantly reduced (P <0 05). After treatment with Dahuang Decoction, the intestinal IgA in both normal and burned mice increased (P < 0.05), and other immune-related substances such as total protein, complement C3, and HDL rhubarb also increased significantly after treatment. Conclusion : The IgA in the intestinal fluid of mice after burn was significantly reduced. Rhubarb has obvious effects on promoting intestinal immune secretion in both normal and burned mice. This effect is one of the important mechanisms of the protective function of rhubarb mucosa barrier.
