目的:研究缩泉丸对肾虚多尿大鼠肾脏MRmRNA及蛋白表达的影响,阐明其“补肾缩尿”的机制。方法:采用腺嘌呤造成大鼠肾虚多尿模型,利用酶联免疫分析法分别检测醛固酮(ALD)的含量;并采用RT-PCR、免疫组化的方法检测各组动物肾脏MRmRNA及蛋白的表达。结果:与模型对照组比较,缩泉丸可显著提高肾虚多尿大鼠血中ALD含量,并可上调模型大鼠肾脏MRm-RNA及蛋白的表达。结论:缩泉丸可通过增加肾脏MRmRNA及蛋白的表达,促进ALD的合成与分泌,从而发挥调节水液代谢的作用。
Objective: To investigate the effect of shrink spring pill on the expression of MR mRNA and protein in kidney of rats with kidney deficiency and to elucidate the mechanism of “kidney shrinking urine”. Methods: Adenine was used to induce kidney-derived polyuria in rats. The levels of aldosterone (ALD) were detected by enzyme-linked immunosorbent assay (ELISA). The mRNA and protein expressions of MRM were detected by RT-PCR and immunohistochemistry. Results: Compared with the model control group, Shouchuwan can significantly increase the ALD level in the kidney and increase the expression of MRm-RNA and protein in the kidney of model rats. Conclusion: Shouchuwan can improve the metabolism of water and water by increasing the expression of MR mRNA and protein in kidney and promoting the synthesis and secretion of ALD.
