目的:观察补肾、健脾、活血三种不同治法对小鼠脾脏BCL-2、Caspase-9 mRNA表达的影响。方法:将雄性NIH小鼠100只,随机分为补肾组、健脾组、活血组、模型组和正常对照组,每组20只。除正常对照组外,分别予以六味地黄汤、补中益气汤、复方丹参饮和生理盐水灌胃给药,并给予环磷酰胺腹腔注射造模,检测脾脏BCL-2、Caspase-9 mRNA表达水平。结果:免疫抑制小鼠BCL-2表达显著降低且Caspase-9表达显著升高。补肾、健脾、活血三方皆能提高BCL-2 mRNA表达,并抑制Caspase-9 mRNA表达(P<0.05)。结论:以六味地黄汤、补中益气汤、复方丹参饮为代表的补肾、健脾、活血三种不同治法能有效地促进BCL-2表达,并抑制Caspase-9表达,这可能是三种不同治法改善环磷酰胺所致免疫抑制小鼠免疫功能的机制之一。
OBJECTIVE: To observe the effects of three different treatments of tonifying the kidney, invigorating the spleen and promoting blood circulation on the expressions of BCL-2 and Caspase-9 mRNA in the spleen of mice. Methods: One hundred male NIH mice were randomly divided into Bushen group, Jianpi group, Huoxue group, model group and normal control group, with 20 rats in each group. In addition to the normal control group, Liuwei Dihuang Decoction, Buzhongyiqi Decoction, Fufang Danshen Decoction and saline were intragastrically administrated, and intraperitoneal injection of cyclophosphamide was given. The expressions of BCL-2 and Caspase-9 mRNA in the spleen Level. Results: The expression of BCL-2 was significantly decreased and the expression of Caspase-9 was significantly increased in immunosuppressed mice. Bushen, invigorating the spleen and promoting blood circulation could increase BCL-2 mRNA expression and inhibit Caspase-9 mRNA expression (P <0.05). Conclusion: The three different treatments of tonifying kidney, invigorating the spleen and activating blood, which are represented by Liuweidihuang decoction, Buzhongyiqi decoction and compound Danshen decoction, can effectively promote the expression of BCL-2 and inhibit the expression of Caspase-9, One of the mechanisms of different treatment methods to improve immune function of immunosuppressed mice induced by cyclophosphamide.
