目的建立三氟甲烷磺酸水解?高效液相色谱(HPLC)测定小麦中结合态脱氧雪腐镰刀菌烯醇(DON)含量的分析方法。方法通过3因素4水平正交试验,确立三氟甲烷磺酸水解小麦中结合态DON的最佳条件。结果水解温度60℃,酸浓度1.0mol/L,时间60min为最佳检测条件。检测了7个小麦样品中的结合态DON含量,在DON污染的小麦样品中均检出有结合态DON,其含量与游离态DON之比为0.20~0.36。结论所建方法具有良好的可靠性和实用性。
OBJECTIVE To establish a method for the determination of bound DON (DON) in wheat by hydrolysis of trifluoromethanesulfonic acid and high performance liquid chromatography (HPLC). Methods The optimum conditions of trifluoro methanesulfonic acid hydrolysis of bound DON in wheat were established by 3-factor 4-level orthogonal experiment. Results The optimum conditions were as follows: hydrolysis temperature 60 ℃, acid concentration 1.0mol / L, time 60min. The content of bound DON in seven wheat samples was detected. The bound DON was detected in DON-contaminated wheat samples, and the content of bound DON was 0.20-0.36 with free DON. Conclusion The method has good reliability and practicability.
