目的:建立高效液相色谱法测定人血浆中尼莫地平浓度的方法。方法:以尼群地平为内标,血浆样品用正己烷-乙醚(1∶1)萃取,采用Diamonsil C18柱(200mm×4.6mm,5μm)分析。流动相为水-甲醇(39∶61),流速1.0mL·min-1,检测波长237nm,柱温35℃。结果:本法在52~1664μg.L-1间线性关系良好(r=0.9996),日内、日间精密度RSD均小于6%,低、中、高浓度的提取回收率均大于95%(n=5)。尼莫地平血浆样品预处理室温放置24h及于-20℃冰箱中贮存15d稳定。结论:本方法简便、快速、准确、重复性好,符合生物样品测定要求,适合尼莫地平药动学的考察。
Objective: To establish a method for the determination of nimodipine in human plasma by high performance liquid chromatography. Methods: Nitrendipine was used as an internal standard. Plasma samples were extracted with n-hexane-ether (1: 1) and analyzed by Diamonsil C18 column (200 mm × 4.6 mm, 5 μm). The mobile phase was water-methanol (39:61), the flow rate was 1.0mL · min-1, the detection wavelength was 237nm and the column temperature was 35 ℃. Results: The method showed good linearity (r = 0.9996) between 52 and 1664μg.L-1, RSD of intra-day and inter-day precision was less than 6%, and the recoveries of low, middle and high concentrations were all above 95% = 5). Nimodipine plasma samples were stored at room temperature for 24h and stored in -20 ℃ refrigerator for 15 days. Conclusion: The method is simple, rapid, accurate and reproducible. It is suitable for the determination of biological samples and is suitable for the study of nimodipine pharmacokinetics.
