目的:研究三七总皂苷(Panax notoginseng saponins,PNS)对快速老化模型小鼠(senescence accelerated mouseprone 8,SAMP 8)脑内突触素(synaptophysin,syp)、tau基因表达的影响。方法:SAMP 8小鼠随机分为PNS低、高(23.38,93.50mg.kg-1.d-1)剂量组、阳性对照药物石杉碱甲(0.038 6 mg.kg-1.d-1)组、模型组。各给药组分别按设定剂量灌胃给药,空白组灌服双蒸水,连续8周。应用实时荧光逆转录聚合酶链式反应(real-time RT-PCR)的方法,检测PNS对SAMP 8脑内syp,tau mRNA含量的影响。结果:PNS高、低剂量组均可以增加syp mRNA含量(P<0.05或P<0.01),PNS对tau mRNA含量未见明显影响。结论:PNS可以在转录水平上调SAMP 8脑内syp基因的表达。
Objective: To investigate the effect of panax notoginseng saponins (PNS) on the expression of synaptophysin (syp) and tau in senescence accelerated mouseprone 8 (SAMP 8) brain. Methods: SAMP 8 mice were randomly divided into low PNS group (23.38, 93.50 mg.kg-1.d-1), positive control group Huperzine A (0.038 6 mg.kg-1.d-1) Group, model group. Each treatment group were given a set dose gavage, blank group doused double distilled water for 8 weeks. The effect of PNS on the content of syp and tau mRNA in SAMP8 brain was detected by real-time RT-PCR. Results: The levels of syp mRNA in both high and low PNS groups were significantly increased (P <0.05 or P <0.01). PNS had no significant effect on tau mRNA. Conclusion: PNS can upregulate the expression of syp gene in SAMP8 brain at the transcriptional level.
