目的:研究健脾复方对人大肠癌移植瘤裸小鼠肿瘤环氧化酶2(COX-2)、核因子κB(NF-κB)和激活物蛋白1(AP-1)表达的影响。方法:将40只近交系(BALB/C)裸小鼠建立人大肠癌原位移植瘤模型,随机分入健脾复方组、塞来昔布组、5-Fu组、模型对照组,另设空白对照组,共分组治疗8周。采用逆转录聚合酶链反应(RT-PCR)方法检测COX-2 mRNA表达,采用免疫组化法检测AP-1和NF-κB的表达。结果:健脾复方组、塞来昔布组和5-Fu组裸小鼠COX-2表达均显著低于模型对照组,且与空白对照组相似(P=0.0019)。健脾复方组、塞来昔布组、5-Fu组和模型对照组AP-1表达高于空白对照组(P=0.0065)。健脾复方组和空白对照组NF-κB表达低于5-Fu组和模型对照组(P<0.05),且健脾复方组与空白对照组间差异未见显著性。结论:健脾复方可以下调COX-2的表达,其机制可能与下调其上游因子NF-κB有关。健脾复方对AP-1表达的影响尚需进一步研究。
Objective: To investigate the effect of Jianpi Compound on the expression of cyclooxygenase 2 (COX-2), nuclear factor κB (NF-κB) and activator protein 1 (AP-1) in human colon carcinoma xenografts in nude mice. Methods: Forty nude mice bearing inbred BALB / C were established in nude mice model of orthotopic colon carcinoma and were randomly divided into three groups: spleen-invigorating compound group, celecoxib group, 5-Fu group and model control group Set a blank control group, a total of 8 weeks treatment group. The expression of COX-2 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR) and the expression of AP-1 and NF-κB was detected by immunohistochemistry. Results: The expressions of COX-2 in spleen-invigorating compound group, celecoxib group and 5-Fu group were significantly lower than those in model control group (P = 0.0019). The expression of AP-1 in spleen-invigorating compound group, celecoxib group, 5-Fu group and model control group was higher than that in blank control group (P = 0.0065). The expression of NF-κB in spleen-invigorating compound group and blank control group was lower than that in 5-Fu group and model control group (P <0.05), and there was no significant difference between Jianpi compound group and blank control group. Conclusion: Jianpi Compound can down-regulate the expression of COX-2, which may be related to the down-regulation of its upstream factor NF-κB. The effect of Jianpi Compound on AP-1 expression needs further study.
