目的:探讨域含蛋白7A(THSD7A)基因在肝细胞癌(HCC)中的表达情况及临床意义。方法:分别用RT-PCR与免疫组化法检测30例新鲜HCC及癌旁组织标本中THSD7A m RNA表达及75例HCC及癌旁组织石蜡标本中THSD7A蛋白的表达,分析其与HCC患者临床病理因素及预后的关系。结果:与癌旁组织比较,在HCC组织中THSD7A m RNA表达下调(P<0.05);THSD7A在HCC组织中的阳性表达率明显低于癌旁组织(24.0%vs.87.0%,P<0.05)。THSD7A蛋白的表达与患者结节数目(P=0.011)、Edmondson-Steiner分级(P=0.013)、BCLC分期(P=0.048)有关。THSD7A蛋白低表达患者总体生存率(P=0.016)与无瘤生存率(P=0.013)均明显低于THSD7A高表达患者。结论:THSD7A在HCC中可能发挥抑癌基因的作用,THSD7A表达降低患者预后不良。
Objective: To investigate the expression of THSD7A gene in hepatocellular carcinoma (HCC) and its clinical significance. Methods: The expression of THSD7A m RNA in 30 specimens of fresh HCC and adjacent non-cancerous tissues was detected by RT-PCR and immunohistochemistry. The expression of THSD7A protein in 75 specimens of HCC and para-cancerous tissues was analyzed. Relationship between factors and prognosis. Results: The expression of THSD7A m RNA in HCC tissues was significantly lower than that in adjacent non-cancerous tissues (P <0.05). The positive rate of THSD7A in HCC tissues was significantly lower than that in adjacent non-cancerous tissues (24.0% vs 87.0%, P <0.05) . The expression of THSD7A protein was correlated with the number of nodules (P = 0.011), Edmondson-Steiner grade (P = 0.013) and BCLC staging (P = 0.048) The overall survival rate (P = 0.016) and disease-free survival rate (P = 0.013) of patients with low THSD7A expression were significantly lower than those with high THSD7A expression. Conclusion: THSD7A may play a role in tumor suppressor gene in HCC, and THSD7A expression may decrease the prognosis of patients.
