目的:观察扶正化瘀方对贮脂细胞胶原生成的影响。方法:①用扶正化瘀方药物血清加入正常传代贮脂细胞中,以正常血清作对照。用~3H—pro掺入、胶原酶消化法计算细胞内、外胶原生成率与胶原生成抑制率。并以PAP法测定细胞内胶原与LN;②DMN造模后用扶正化瘀方体内给药,生理盐水作对照,连续7天后处死.分离细胞,以同样方法计算纤维肝原代贮脂细胞细胞内、外胶原生成率与胶原生成抑制率。结果:①药物血清能明显减少正常传代贮脂细胞细胞外胶原生成率,抑制细胞Ⅰ、Ⅲ、Ⅳ型胶原与LN的表达;②体内给药后DMN纤维肝原代贮脂细胞细胞内、外胶原生成率均明显抑制。结论:扶正化瘀方能抑制贮脂细胞的胶原合成。
Objective: To observe the effect of Fuzheng Huayu Decoction on the collagen production of fat-storing cells. Methods: 1 Fuzheng Huayufang drug serum was added into normal passage lipid-storing cells and normal serum was used as a control. Using 3H-pro incorporation and collagenase digestion method, the intracellular and extracellular collagen production rate and inhibition rate of collagen production were calculated. The PAP method was used to measure intracellular collagen and LN; after 2DMN was modeled, it was treated with Fuzheng Huayufang in vivo and physiological saline was used as a control. After being sacrificed continuously for 7 days, the cells were detached and the cells were calculated by the same method. The rate of collagen production and collagen production inhibition rate. RESULTS: 1The drug serum can significantly reduce the extracellular collagen production rate of normal passage fat-storing cells, inhibit the expression of type I, III, IV collagen and LN; 2 DMN fiber liver primary fat-storing cells intracellularly and externally after in vivo administration Collagen production rate was significantly inhibited. Conclusion: Fuzheng Huayu Decoction can inhibit collagen synthesis in fat-storing cells.
