探讨醋酸铅对脐带间质干细胞生物学特性的影响。用细胞计数法测定脐带间质干细胞活性,流式细胞仪检测醋酸铅对脐带间质干细胞凋亡的影响,用ALP组织染色分析成骨诱导的变化,并用RT-PCR和定量PCR分析醋酸铅作用前后的细胞因子表达情况。结果显示:脐带间质干细胞活力随醋酸铅浓度的升高而受抑,60μmol/L的醋酸铅可引起脐带间质干细胞凋亡率增加;成骨诱导分化后ALP表达率下降,TPO、SCF和VEGF等细胞因子表达也有所下降。因此,醋酸铅可以抑制脐带间质干细胞的增殖、促进其凋亡并影响其多向分化潜能。
To investigate the effect of lead acetate on the biological characteristics of umbilical cord mesenchymal stem cells. Umbilical cord mesenchymal stem cell activity was assayed by cell counting method. Apoptosis of umbilical cord mesenchymal stem cells was detected by flow cytometry. Osteoinductive changes were detected by ALP staining. RT-PCR and quantitative PCR were used to analyze the effect of lead acetate Cytokine expression. The results showed that: umbilical cord mesenchymal stem cell viability was inhibited with the increase of lead acetate concentration, 60μmol / L lead acetate can lead to increased apoptosis rate of umbilical cord mesenchymal stem cells; osteogenic induction differentiation ALP expression decreased, TPO, SCF and VEGF Cytokine expression also declined. Therefore, lead acetate can inhibit the proliferation of umbilical cord mesenchymal stem cells, promote its apoptosis and affect its multi-directional differentiation potential.
