目的探讨采用GDNF基因转染大鼠周围源性OECs,并对GDNF基因转染的周围源性OECs表达GDFN量及稳定程度进行测定和分析,以期为脊髓损伤的基因治疗提供一定依据。方法提取SD大鼠的周围源性OECs,采用重组腺病毒作为载体,转染GDFN到大鼠周围源性OECs,并采用生物学方法测定转染细胞GDFN的表达水平。结果 GDFN成功转染至大鼠周围源性OECs中,转染率为77%,转染细胞的GDFN分泌量为30.3ng/106/24h,并可稳定表达。结论将GDNF作为目的基因转染周围源性OECs进行SCI的基因治疗的是合理的、可行的。
Objective To investigate the expression of GDFN in peripheral OECs of GDNF gene transfected by GDNF gene and the degree of stabilization of GDFF gene in peripheral blood of rats with GDNF gene transfection and to provide some evidences for gene therapy of spinal cord injury. Methods The peripheral OECs of SD rats were extracted and transfected into peripheral OECs of rats with recombinant adenovirus using GDFN as a vector. The expression of GDFN in the transfected cells was determined by biological methods. Results GDFN was successfully transfected into peripheral OECs of rats. The transfection rate was 77%. The GDFN secretion of the transfected cells was 30.3ng / 106 / 24h and was stable. Conclusion It is reasonable and feasible to use GDNF as the target gene to transfect peripheral OECs for gene therapy of SCI.
