目的:对三草方水提取物联合拓扑替康进行抗小肺癌肿瘤活性研究。方法:分别采用插块法和细胞接种法建立移植人小细胞肺癌H446的裸鼠模型,观察不同剂量及不同组合三草方水提取物联合拓扑替康抗小细胞肺癌肿瘤的效果。结果:三草方水提取物100mg/kg剂量组效果优于其他剂量三草方水提取物;在插块法建立肿瘤裸鼠皮下移植模型中,拓扑替康1.5mg/kg剂量组的T/C(%)为29.8%,三草方水提取物联合用药组的T/C(%)分别为20.1%和16.5%,拓扑替康组和联合用药组的肿瘤增长率、肿瘤体积与空白组比较有显著差异(P<0.01);在细胞接种法建立肿瘤裸鼠皮下移植模型中,拓扑替康1.5mg/kg剂量组的T/C(%)为30.5%,三草方水提取物不同剂量与拓扑替康联合用药组的T/C(%)分别为24.2%和15.2%,联合用药组的肿瘤增长率、肿瘤体积与对照比较有显著差异,表现显著的抑制肿瘤生长的作用。结论:三草方水提取物(100mg/kg)与拓扑替康(0.75mg/kg)联合,在抗小细胞肺癌动物模型中表现出一定的协同作用。
OBJECTIVE: To study the antitumor activity of Sancao Fang water extract combined with topotecan in small lung cancer. Methods: The nude mouse model of H446 human small cell lung cancer was established by inserting block method and cell inoculation method. The effect of different doses and combination of three herbs and water extract in combination with topical on anti-small cell lung cancer was observed. Results: The water extract of Sancao Fang 100mg / kg dose group was superior to the other doses of Sancaofan water extract; insert nude mice established subcutaneous transplantation model, topotecan 1.5mg / kg dose group T / The C (%) was 29.8%. The T / C (%) of Sancao Fang water extract combination group was 20.1% and 16.5% respectively. The tumor growth rate and tumor volume in topotecan group and combination group were significantly lower than those in blank group (P <0.01). The T / C (%) of topotecan 1.5mg / kg dose group was 30.5% in the model of subcutaneous transplantation of tumor inoculated by cell inoculation method, and the water extract of Sancao Fang was different T / C (%) was 24.2% and 15.2% for the combination of topiramate and topotecan, respectively. The tumor growth rate and tumor volume in the combination group were significantly different from those in the control group, indicating significant inhibition of tumor growth. Conclusion: The combination of Sancao Fang water extract (100mg / kg) and topotecan (0.75mg / kg) has synergistic effect in the animal model of NSCLC.
