The FAM3 family consists of 4 members: FAM3 / B / C / D. FAM3A expression is down-regulated in diabetic mice and human livers. Overexpression of FAM3A in the liver of diabetic mice activated Akt and inhibited FOXO1 activity, relieving hyperglycemia, insulin resistance and fatty liver. FAM3A protein is localized to the mitochondria and promotes ATP synthesis and secretion. Secreted ATP activates Akt signaling through the P2 receptor and regulates hepatocyte glycolipid metabolism.
