目的:研究胚胎干细胞相关基因Oct4和Nanog在人胰腺癌肿瘤干细胞和普通肿瘤细胞中表达的差异。方法:用流式细胞仪在人胰腺癌Panc-1细胞系中分选肿瘤干细胞,细胞免疫荧光法检测2种肿瘤干细胞和Panc-1细胞中Oct4、Nanog基因的表达情况。结果:Oct4及Nanog基因在2种分选出的细胞中表达均高于未分选细胞。结论:干细胞相关基因Oct4、Nanog在胰腺癌Panc-1肿瘤干细胞中的表达高于普通胰腺癌细胞。
OBJECTIVE: To study the differences of expression of embryonic stem cell related genes Oct4 and Nanog in human pancreatic cancer stem cells and normal tumor cells. METHODS: Tumor stem cells were sorted by flow cytometry in Panc-1 human pancreatic cancer cell lines. The expression of Oct4 and Nanog genes in two tumor stem cells and Panc-1 cells was detected by immunofluorescence. Results: The expression of Oct4 and Nanog genes were higher in both sorted cells than in unsorted cells. Conclusion: The expression of stem cell related genes Oct4 and Nanog in pancreatic cancer Panc-1 tumor stem cells is higher than that in normal pancreatic cancer cells.
