目的:采用石油醚萃取、大孔树脂及凝胶树脂吸附、活性炭超声、RP-18层析4种方法进行研究与比较,筛选出益母草提取液的最佳脱色除杂方法。方法:以益母草中主要有效成分益母草碱、芦丁、金丝桃苷及总固型物为考察指标,考察不同方法的脱色除杂效果。结果:益母草提取液经不同方法脱色后,总固型物的保留率:活性炭>MCI gel>D101>RP-18>Diaion HP 20。若以3倍于药材量的RP-18为吸附剂,80%甲醇为洗脱剂,对益母草提取物脱色后,总有效成分(以益母草碱、芦丁、金丝桃苷计)保留率最高。结论:RP-18对益母草提取物中的色素具有较好的吸附性能,适于除杂纯化。
OBJECTIVE: To study and compare the four methods of petroleum ether extraction, macroporous resin and gel resin adsorption, activated carbon ultrasonography and RP-18 chromatography to screen out the best bleaching and impurity removal methods of Leonurus extract. Methods: Leonurine, rutin, hyperoside and total solid were the main active ingredients in Leonurus. The effects of different methods on decolorization and impurity removal were investigated. Results: After the mother liquor extracts were decolorized by different methods, the retention rate of total solid was: activated carbon> MCI gel> D101> RP-18> Diaion HP 20. If 3 times the amount of crude drug RP-18 as adsorbent, 80% methanol as eluent, after the motherwort extract decolorization, the total active ingredient (Leonurine, rutin, hyperoside dollars) the highest retention rate . Conclusion: RP-18 has good adsorption properties on the pigment of Motherwort extract, suitable for purifying.
