目的 :了解RhD(- )中国汉族人RHD基因的构成情况 ,并探讨其潜在的应用价值。方法 :采用PCR SSP方法分析 5 0名RhD(+)和 76名常规血清学RhD(- )汉族供血者的RHD基因存在情况 ,对其中 8例RhD(- )血样进行了吸收放散试验。结果 :5 0名RhD(+)供者RHD基因完整存在。 76名RhD(- )供者中 ,2 2名供者(2 8 95 % )存在完整RHD基因 ,9名供者 (11 84% )缺失大部分RHD基因 ,并全为中间缺失型 ;45名供者(5 9 2 1% )完整缺失RHD基因。携带完整RHD基因片段的RhD(- )个体表型均为CC或Cc,而无cc。吸收放散试验证实携带完整RHD基因的常规血清学RhD(- )个体中存在吸收放散弱D型 (Delution ,Del)。结论 :常规血清学RhD(- )中国人RHD基因存在多态性 ,提示中国汉族人RhD(- )特征有多种遗传机制 ,因此在临床医学中要正确对待不同遗传背景的常规血清学RhD(- )个体。
Objective: To understand the composition of RhD (-) Chinese Han RHD gene and to explore its potential value. Methods: The presence of RHD gene in 50 donors of RhD (+) and 76 conventional RhD (-) Han donors was analyzed by PCR SSP method, and 8 cases of RhD (-) blood samples were tested for absorption and desorption. Results: RHD gene was completely existed in 50 RhD (+) donors. Of the 76 RhD (-) donors, a complete RHD gene was present in 2 of 22 donors (2,895%), and most RHD genes were deleted in 9 donors (11,884% The donor (5 921%) completely deleted the RHD gene. The RhD (-) individuals carrying the complete RHD gene were phenotypically CC or Cc with no cc. Absorption and desorption assays demonstrated the presence of Delution (Del) in conventional serological RhD (-) individuals carrying the complete RHD gene. CONCLUSIONS: Conventional serology RhD (-) RHD gene polymorphism in Chinese suggests that there are multiple genetic mechanisms of RhD (-) in Han Chinese. Therefore, it is necessary to correctly treat the common serological RhD (- -) Individuals.
