目的:研究坤泰胶囊(KTC)对SD大鼠实验性子宫异位内膜生长及其妊娠结局的影响。方法:手术法建立SD大鼠子宫内膜异位症(EMs)模型。将造模成功的SD大鼠分为模型对照组、阳性对照组(每只给予醋酸亮丙瑞林0.1 mg皮下注射,共1次),KTC低剂量组(每只按0.24 g/kg体质量灌胃,qd×21 d)、KTC高剂量组(每只按0.60 g/kg体质量灌胃,qd×21 d),每组14只。给药21 d后,每组解剖8只,比较给药前、后异位内膜体积变化;Western blotting法测定异位内膜caspase-8、caspase-3和caspase-9蛋白表达。每组剩余6只与雄鼠交配,妊娠12 d后,解剖观察受孕情况,并对胎盘、胎仔称量。动情周期正常的SD大鼠14只作为空白对照。结果:与模型对照组相比,KTC低、高剂量组体质量无统计学差异(P>0.05);KTC低、高剂量组异位内膜抑制率分别为(40.25±35.18)%和(65.52±15.82)%;KTC低、高剂量组异位内膜中caspase-8、caspase-9和活化caspase-3明显升高(P<0.05);KTC低剂量组的大鼠胎仔数量呈增加趋势。结论:KTC通过升高caspase-8、caspase-9和活化caspase-3蛋白含量,促进异位内膜的凋亡;KTC低剂量组EMs大鼠受孕率呈现出升高的趋势。
Objective: To study the effect of kun tai capsule (KTC) on experimental uterine ectopic endometrial growth and pregnancy outcome in SD rats. Methods: SD rat model of endometriosis (EMs) was established by surgery. The successful SD rats were divided into model control group, positive control group (each given 0.1 mg leuprolide 0.1 mg subcutaneously once), KTC low dose group (each dose of 0.24 g / kg body mass Gd × 21 d), KTC high dose group (each gavage at 0.60 g / kg body weight, qd × 21 d), 14 rats in each group. After 21 days of administration, 8 mice in each group were dissected to compare ectopic endometrium volume changes before and after administration. The protein expressions of caspase-8, caspase-3 and caspase-9 in ectopic endometrium were detected by Western blotting. The remaining 6 mice in each group were mated with the male rats. After 12 days of gestation, the conceptions were observed and the placenta and fetus were weighed. 14 rats with normal estrous cycle as a blank control. Results: Compared with the model control group, there was no significant difference in the body weight of KTC low and high dose groups (P> 0.05). The inhibition rates of ectopic endometrium in KTC low and high dose groups were (40.25 ± 35.18)% and (65.52 ± 15.82)%. The levels of caspase-8, caspase-9 and activated caspase-3 in ectopic endometrium of KTC low and high dose groups were significantly increased (P <0.05). CONCLUSION: KTC promotes the apoptosis of ectopic endometrium by increasing the contents of caspase-8, caspase-9 and activated caspase-3. The pregnancy rate of EMs rats in KTC low dose group shows an increasing trend.
