目的:探讨Ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid(5F)对人绒癌JAR细胞侵袭、转移、黏附的影响及MMP-9、VEGF表达的变化。方法:用不同浓度的5F作用JAR细胞,MTT比色法,Transwell小室,细胞黏附实验观察其增殖、侵袭、迁移、黏附能力的变化;Western blot方法检测5F对JAR细胞MMP-9、VEGF表达水平的影响。结果:作用6 h,5F能抑制JAR细胞的增殖、转移、侵袭、黏附,其抑制率呈剂量效应(P<0.05)。5F作用24 h JAR细胞,MMP-9、VEGF蛋白表达水平能明显下调。结论:5F能抑制JAR细胞的侵袭、转移、黏附并下调MMP-9、VEGF蛋白表达水平。
Objective: To investigate the effect of Ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid on the invasion, metastasis and adhesion of human cervical carcinoma JAR cells and the changes of MMP- Methods: The proliferation, invasion, migration and adhesion of JAR cells were detected by different concentrations of 5F JAR cells, MTT assay, Transwell chamber and cell adhesion assay. The expression of MMP-9 and VEGF in JAR cells was detected by Western blot Impact. Results: After 6 h treatment, 5F inhibited the proliferation, metastasis, invasion and adhesion of JAR cells in a dose-dependent manner (P <0.05). The expression of MMP-9 and VEGF in JAR cells induced by 5F for 24 h was significantly down-regulated. CONCLUSION: 5F can inhibit the invasion, metastasis, adhesion and down-regulate the expression of MMP-9 and VEGF in JAR cells.
