目的研究脑出血大鼠延髓内脏带(MVZ)内孤束核、迷走神经背核FOS蛋白表达变化规律,探讨MVZ在脑源性多器官功能障碍综合征(MODS)的神经免疫调节作用。方法(1)向大鼠尾状核内注射Ⅶ型胶原酶0.8 U建立脑出血模型,54只Wistar大鼠随机被分为对照组、假手术组及出血后4、8、12、24、36、48和72 h时相点的7个亚组;(2)应用免疫组织化学ABC法观察各时相点MVZ内孤束核、迷走神经背核FOS蛋白的表达变化规律。结果(1)在对照组和假手术组MVZ内孤束核、迷走神经背核FOS蛋白阳性表达极少;出血组FOS蛋白阳性细胞在孤束核、迷走神经背核密集表达,24~36 h达高峰,在72 h仍有阳性表达。结论孤束核、迷走神经背核FOS蛋白的阳性表达增多提示MVZ是脑出血致MODS的神经免疫调节中枢之一。
Objective To investigate the changes of FOS protein expression in the nucleus tractus solitarius and the dorsal vagal nucleus in the medullary visceral zone (MVZ) of rats with intracerebral hemorrhage and to explore the neuroimmunologic regulation of MVZ in brain-derived multiple organ dysfunction syndrome (MODS). Methods (1) Intrathecal injection of type Ⅶ collagenase 0.8 U into the caudate nucleus of rats was used to establish intracerebral hemorrhage model. Fifty-four Wistar rats were randomly divided into control group, sham operation group and 4, 8, 12, 24, 36 , 48h and 72h respectively. (2) The expression of FOS protein in nucleus tractus solitarius and vagus dorsal nucleus of MVZ in each time point was observed by immunohistochemical ABC method. Results (1) In the control group and sham operation group, the positive expression of FOS protein in nucleus tractus solitarius and vagus dorsal nucleus in MVZ was very few. The expression of FOS protein positive cells in the hemorrhage group was densely expressed in nucleus tractus solitarius and vagal nucleus and peaked at 24- 36 h , There is still positive expression at 72 h. Conclusion The increased expression of FOS protein in nucleus tractus solitarius and vagus dorsal vagal nucleus suggests that MVZ is one of the neuroimmunoregulatory centers of MODS induced by intracerebral hemorrhage.
