目的:建立射干中芒果苷的含量测定方法;测定不同产地射干中芒果苷的含量。方法:采用RP-HPLC法,外标法定量,Kromail C18柱分离,流动相:乙腈-0.2%磷酸水溶液(15∶85),检测波长257 nm,柱温为25℃,流速1mL.min-1。从湖北、安国、北京及沈阳分别采集射干供试品;对湖北及安国射干根茎与须根中芒果苷的含量进行比较。结果:所建立的RP-HPLC含量测定方法线性关系良好(r>0.9990),方法平均回收率98.65%,RSD=1.62%(n=5)。结论:所建立的RP-HPLC含测方法简单,快速,专属性强好。不同产地射干中芒果苷的含量有差异,须根中芒果苷含量显著高于根茎中。
Objective: To establish a method for the determination of mangiferin in dried shoots and to determine the content of mangiferin in dried shoots from different areas. Methods: RP-HPLC method was used for the quantitative determination by external standard method on a Kromail C18 column. The mobile phase was acetonitrile-0.2% phosphoric acid solution (15:85), the detection wavelength was 257 nm, the column temperature was 25 ℃ and the flow rate was 1 mL.min-1 . The test samples were collected from Hubei, Anguo, Beijing and Shenyang respectively. The contents of mangiferin in stems and roots of Shegan from Hubei and Anguo were compared. Results: The established RP-HPLC method was linear (r> 0.9990). The average recovery was 98.65% with RSD = 1.62% (n = 5). Conclusion: The established RP-HPLC method is simple, rapid and specific. There were differences in the content of mangiferin in the stems and shoots in different areas. The content of mangiferin in the fibrous roots was significantly higher than that in the rhizomes.
