目的检测不同化疗敏感程度的卵巢上皮癌组织中P53基因的表达,分析其与化疗耐药的关系。方法 80例新鲜卵巢上皮癌组织均由手术中取得,采用ATP-TCA技术检测卵巢癌组织对八种化疗药物的体外敏感性。采用免疫组化SP法检测80例组织中P53基因的表达情况。结果对多西他赛化疗耐药的卵巢癌组织中,P53基因表达阳性率显著高于化疗敏感的组织(P=0.003),在对紫杉醇、卡铂及依托泊苷耐药的组织中其表达阳性率也显著升高(P<0.05)。结论研究结果提示:P53基因的高表达与卵巢癌化疗耐药相关,P53基因可能作为卵巢上皮癌化疗耐药性的预测指标。
Objective To detect the expression of P53 gene in ovarian epithelial carcinoma with different chemosensitivity and to analyze its relationship with chemoresistance. Methods 80 cases of fresh epithelial ovarian cancer tissues were obtained by surgery. ATP-TCA was used to detect the sensitivity of ovarian cancer tissues to eight chemotherapeutic drugs in vitro. Immunohistochemical SP method was used to detect the expression of P53 gene in 80 cases. Results The positive rate of P53 gene expression in docetaxel - resistant ovarian cancer tissues was significantly higher than that in chemosensitive tissues (P = 0.003). The expression of P53 gene in paclitaxel, carboplatin and etoposide resistant tissues The positive rate was also significantly increased (P <0.05). Conclusion The results suggest that high expression of P53 gene is associated with chemoresistance in ovarian cancer, and P53 gene may be used as a predictor of chemoresistance in ovarian epithelial cancer.
