目的阐明2012年邯郸市分离的Victoria系乙型流感毒株HA1抗原性和基因变异特征。方法对2012-2013年分离的流感病毒采用RT-PCR方法特异性扩增HA1基因,扩增产物进行核苷酸序列测定,用DNAStar中Megalign软件进行比对分析,构建系统进化树。结果邯郸市2012年分离的Victoria系乙型流感病毒与WHO推荐的2009-2012年北半球代表株相比,核苷酸同源性为98.1%~99.2%,氨基酸同源性为98.2%~99.4%。HA1区未发生氨基酸丢失和插入,与B/Brisbane/60/2008相比2个位点氨基酸替换具有共性(146I>V和197D>N),而这2个位点与2006-2008年疫苗株B/Malaysia/2506/04相同。与B/Brisbane/60/2008相比7个毒株中有2个毒株发生了另外4个位点(58L>P、129N>S、171N>D和174G>E)的替换。结论 2012年邯郸市分离的Victoria系乙型流感病毒血凝素蛋白未发生明显的抗原性改变,与WHO推荐的2011-2012年的流感疫苗株B/Brisbane/60/2008匹配,不属于Victoria系乙型流感病毒新变种。
Objective To elucidate the antigenicity and genetic variation of HA1 isolates of influenza B virus isolated in Handan City in 2012. Methods The HA1 gene was amplified by RT-PCR from influenza virus isolated from 2012 to 2013. The amplified product was sequenced and compared with DNAStar Megalign software to construct the phylogenetic tree. Results Compared with the representative strains of the Northern Hemisphere from 2009 to 2012 recommended by WHO, the isolates of Victoria-type B influenza virus isolated in 2012 in Handan City showed nucleotide homology of 98.1% -99.2% and amino acid homology of 98.2% -99.4% . There were no amino acid deletions and insertions in the HA1 region. The amino acid substitutions at the two sites were common (146I> V and 197D> N) compared to B / Brisbane / 60/2008, and these two sites were similar to the 2006-2008 vaccine strain B / Malaysia / 2506/04 the same. Two of the seven strains were replaced by another four loci (58L> P, 129N> S, 171N> D and 174G> E) compared to B / Brisbane / 60/2008. Conclusions There was no significant antigenic change in the influenza B virus isolated from Handan City in 2012. It matches with the WHO recommended influenza vaccine strain B / Brisbane / 60/2008 from 2011 to 2012 and is not a member of the Victoria family A new variant of influenza B virus.
