目的制备盐酸万乃洛韦微孔渗透泵片,考察该制剂的释药影响因素及释药机理。方法利用正交设计优化处方,根据不同时间累积释放度考察药物的释放情况。结果盐酸万乃洛韦微孔渗透泵控释片体外释药速度较市售片缓慢、平稳。结论本制剂释药速率随渗透压差减小而成比例减小,遵从以渗透压差为释药动力的释药模式,12 h内呈现良好的零级释放特征。
Objective To prepare a micro-porous osmotic pump tablet of valacyclovir hydrochloride and investigate its influencing factors and drug release mechanism. Methods Orthogonal design optimization prescription, according to the cumulative release of different time to investigate the release of drugs. Results Valacyclovir hydrochloride microporous osmotic pump controlled release tablets in vitro release rate than the slow, steady. Conclusion The drug release rate decreases proportionally with the decrease of osmotic pressure, and follows the mode of drug release with osmotic pressure difference as the drug releasing power, which shows good zero-order release characteristics within 12 h.
