目的通过体外细胞实验探讨建中理劳汤含药血清对TGF-β1诱导大鼠系膜细胞HBZY-1纤维化过程中TGF-β/Smad信号通路的调控作用。方法采用PCR、Western Blot方法观察建中理劳汤含药血清对TGF-β1诱导大鼠系膜细胞HBZY-1纤维化过程中Smad-2、Smad-3、Smad-7 mRNA及蛋白表达的影响;通过Western Blot方法检测5.5μg/L的TGF-β1诱导HBZY-1细胞不同时间p-Smad2、p-Smad3蛋白的表达影响;Western Blot检测建中理劳汤大鼠含药血清对TGF-β1诱导30min后的HBZY-1细胞p-Smad2、p-Smad3蛋白的表达影响。结果与正常组相比,TGF-β1组中p-Smad2、p-Smad3蛋白表达显著升高,Smad-7表达明显降低(P<0.05);而与TGF-β1组相比,加入建中理劳汤含药血清后,p-Smad2、p-Smad3蛋白表达显著降低,Smad-7表达明显升高(P<0.05)。结论建中理劳汤含药血清对TGF-β1诱导大鼠系膜细胞HBZY-1纤维化过程中TGF-β/Smad信号通路起调控作用,可能是其抗纤维化形成的机制之一。
Objective To investigate the regulatory effect of Jianzhonglilao containing serum on TGF-β / Smad signaling pathway in rat mesangial cell line HBZY-1 induced by TGF-β1 in vitro. Methods The mRNA and protein expressions of Smad-2, Smad-3 and Smad-7 in TGF-β1-induced fibrosis of rat mesangial cells HBzY-1 were detected by PCR and Western Blot The expression of p-Smad2 and p-Smad3 in HBZY-1 cells induced by 5.5μg / L TGF-β1 was detected by Western Blot. Western Blot was used to detect the expression of TGF-β1 The expression of p-Smad2 and p-Smad3 in HBZY-1 cells induced 30min after induction. Results Compared with the normal group, the expressions of p-Smad2 and p-Smad3 in TGF-β1 group were significantly increased and the expression of Smad-7 was significantly decreased (P <0.05). Compared with TGF-β1 group, After treatment with serum containing Louroup, the expressions of p-Smad2 and p-Smad3 protein were significantly decreased and Smad-7 expression was significantly increased (P <0.05). Conclusion Jianzhonglilao containing serum can regulate TGF-β / Smad signaling pathway in TGF-β1-induced fibrosis of rat mesangial cells and may be one of the mechanisms of anti-fibrosis.
