目的:考察水解酪蛋白粉(Casein hydrolysate,CH)对天麻、钩藤和杜仲复方(Chines herbal compound,CHC)降血压作用的影响和急性毒性。方法:将SD大鼠分为空白对照组和样品正常组,给予蒸馏水和CH与CHC配伍药物;将原发性高血压大鼠(SHR)分为牛黄降压胶囊组、CH-CHC药物低、中、高剂量组、CH组和CHC组及模型对照组,每天灌胃给药1次,连续4周,记录大鼠收缩压、舒张压、心率和血清一氧化氮(NO)、血管紧张素Ⅱ(AngⅡ)、内皮素-1(ET-1)含量;急性毒性实验以KM小鼠按最大给药体积灌胃,每天2次,连续14天,记录动物一般变化和主要脏器系数。结果:与模型对照组比较,CH-CHC中高剂量组、CH组和CHC组大鼠收缩压和舒张压显著降低(P<0.05或P<0.01),CH-CHC降血压作用显著优于CH组和CHC组(P<0.05),相应血清AngⅡ、NO和ET-1水平也具有显著性差异,各药物组对大鼠心率均无显著影响,急性毒性试验药物组与对照组结果无显著差异。结论:CH可以显著增强天麻、钩藤和杜仲复方的降血压作用,两者配伍无急性毒性作用。
Objective: To investigate the effects of hydrolyzed casein hydrolysate (CH) on the hypotensive effect of Gastrodia elata Blume, Uncaria rhynchophylla and Chines herbal compound (CHC) and its acute toxicity. Methods: The SD rats were divided into blank control group and normal sample group, given distilled water and CH and CHC compatibility drugs; the SHR was divided into Niuhuang Jiangya Capsule group, CH-CHC low drug, Middle and high dose group, CH group and CHC group and model control group were given gavage once daily for 4 weeks. Systolic blood pressure, diastolic blood pressure, heart rate and serum nitric oxide (NO), angiotensin (AngⅡ) and endothelin-1 (ET-1). Acute toxicity test KM mice were intragastrically administrated twice a day for 14 consecutive days, and the general changes and major organ coefficients of animals were recorded. Results: Compared with the model control group, the CH-CHC middle-high dose group, CH group and CHC group systolic pressure and diastolic pressure were significantly lower (P <0.05 or P <0.01), CH-CHC hypotensive effect was significantly better than the CH group And CHC group (P <0.05). The corresponding serum AngⅡ, NO and ET-1 levels also had significant differences. Each drug group had no significant effect on the heart rate of rats. There was no significant difference between the acute toxicity test drug group and the control group. Conclusion: CH can significantly enhance the antihypertensive effects of Gastrodia elata Blume, Uncaria rhynchophylla and Eucommia ulmoides complex, and the combination of them has no acute toxicity.
