目的建立Balb/c小鼠单纯心理应激模型及其评价体系,观察单纯心理应激对机体内分泌免疫功能的影响。方法采用旁观电击实验建立心理应激模型,20只Balb/c小鼠随机分为正常对照组10只和心理应激组10只,另外准备10只小鼠仅接受足部电击,作为应激源,不进入实验处理。记录并分析正常对照组和心理应激组小鼠行为学指标,检测血清皮质酮水平,血清IL-2、IL-4含量,评价Balb/c小鼠单纯心理应激模型。结果心理应激组小鼠旷场试验指标、血清IL-2水平较对照组有显著降低(P<0.05);血清皮质酮、IL-4水平较对照组有显著增高(P<0.05)。结论单纯心理应激小鼠模型建立成功,该心理应激模型能激活下丘脑-垂体-肾上腺轴,抑制小鼠的细胞免疫功能。
Objective To establish a model of pure psychological stress in Balb / c mice and its evaluation system and observe the effect of pure psychological stress on the endocrine and immune function of the body. Methods A model of psychophysical stress was established by observing shock waves. Twenty Balb / c mice were randomly divided into normal control group (n = 10) and psychological stress group (n = 10). Ten additional mice were given foot shock only as stress source , Do not enter the experimental treatment. The behavioral indexes of normal control group and psychological stress group were recorded and analyzed. Serum corticosterone level, serum IL-2 and IL-4 levels were measured to evaluate the simple psychological stress model of Balb / c mice. Results Compared with control group, the level of serum IL-2 in the psychological stress group was significantly lower than that in the control group (P <0.05). The levels of serum corticosterone and IL-4 were significantly increased (P <0.05). Conclusion The simple psychological stress mouse model was successfully established. The psychological stress model could activate the hypothalamus - pituitary - adrenal axis and inhibit the cellular immune function in mice.
