目的:探究大黄庶虫虫丸对气虚血瘀大鼠炎性因子TNF-α和NF-kB/IkB-α信号通路的影响。方法:采用游泳力竭联合注射盐酸肾上腺素复合因素致气虚血瘀的动物模型。大黄庶虫虫丸灌胃干预治疗,造模1周,注射肾上腺素1 h后麻醉,股动脉取血,检测血流变学,并采用放射免疫法测定TNF-α水平, ABC免疫组化法测定NF-kB、 IkB-α信号通路的蛋白表达。结果:模型组与空白组比较TNF-α、 NF-kB显著升高, IkB-α表达明显降低。大黄庶虫虫丸高剂量组与模型组比较, TNF-α、 NF-kB表达明显降低, IkB-α表达明显增高。结论:大黄庶虫虫丸能够降低气虚血瘀大鼠炎性因子TNF-α表达,并通过IkB-α抑制NF-kB的表达进一步抑制炎症的发生和发展,发挥祛瘀补虚之功效。“,”Objective:to explore the effect of Dahuang Zhechong Wan on TNF-αand NF-kB/IkB-αin rats with qi deficiency and blood stasis.Methods:animal models with qi deficiency and blood stasis were induced by the exhausting swimming combined with in-jection of epinephrine hydrochloride.Dahuang Zhechong Wan was taken by gavage for intervention treatment.One week after the es-tablishment, the rats were injected with epinephrine hydrochloride, and anesthetized one hour later.Blood was taken from femoral ar-tery to determine the blood rheology, TNF-αby radio-immunity, and the protein expression of NF-kB/IkB-αby ABC Immunohisto-chemisty.Results:compared to control group, TNF-αand NF-kB of model group increased significantly, and IkB-αdecreased signifi-cantly.Compared to model group, TNF-αand NF-kB of high dose group decreased sighnificantly, and IkB-αincreased significantly. Conclusion:Dahuang Zhechong Wan can decrease TNF-αexpression in rats with qi deficiency and blood stasis, and inhibit NF-kB through IkB-αso to further inhibit the occurrence and development of inflammation and play the role of removing blood stasis and rein-forcing deficiency.
