目的 运用慢性移植物抗宿主反应 (GVHR)诱导系统性红斑狼疮 (SLE)样小鼠模型 ,为进一步探索SLE发病机制奠定基础。方法 将亲代BALB/C小鼠淋巴细胞经静脉途径输入 (BALB/C×C57BL/ 6 )F1代小鼠体内 ,用ELISA测定IgG类抗dsDNA抗体和抗组蛋白抗体 ,用免疫荧光法检测抗核抗体 (ANA)核型和免疫复合物沉积 ,用免疫印迹法测定抗可浸出核抗原 (ENA)抗体。结果 亲代淋巴细胞所致的慢性移植物抗宿主反应可诱导F1代小鼠产生高滴度的抗dsDNA抗体、抗组蛋白抗体等多种ANA ,免疫小鼠的肾脏有显著的免疫复合物沉着。结论 运用慢性GVHR诱导的SLE样小鼠模型 ,小鼠发病迅速 ,抗核抗体的产生与人SLE相似 ,有利于实验室进行有关SLE的研究
Objective To induce systemic lupus erythematosus (SLE) -like mouse model by chronic graft-versus-host reaction (GVHR), and lay the foundation for further exploration of the pathogenesis of SLE. Methods The BALB / C mouse lymphocytes were transplanted into the F1 generation of BALB / C mice via intravenous route. IgG anti-dsDNA antibodies and anti-histone antibodies were detected by ELISA. The anti-nuclear antibodies were detected by immunofluorescence Antibody (ANA) karyotype and immune complexes were deposited and anti-leachable nuclear antigen (ENA) antibodies were determined by immunoblotting. Results The chronic graft-versus-host reaction induced by the parental lymphocytes induced a high titer of anti-dsDNA antibody and anti-histone antibody and other ANA in F1 generation mice. The kidneys of immunized mice had significant immunocomplex deposition. Conclusion The SLE-like mouse model induced by chronic GVHR shows rapid onset in mice and the production of antinuclear antibodies is similar to that of human SLE, which is conducive to laboratory studies on SLE
