建立了 RP- HPLC法测定水飞蓟宾在人血浆中浓度的方法。用乙腈沉淀血浆蛋白 ,离心后 ,取上清液 10 μl进样分析。色谱条件 :C18柱 (10 μm) 2 2 0 mm× 4 .6 mm;流动相为 :V(甲醇 )∶ V(水 )∶ V(0 .5mol· L-1磷酸二氢钾 ) =10∶ 10∶ 1(用 0 .2 mol· L-1磷酸调 p H=4 .0 ) ;流速为 1.0 ml· min-1;检测波长为 2 90 nm。结果水飞蓟宾 A、B平均回收率分别为 98.96 %、97.93% ;日内 RSD分别为3.0 8%、2 .15% ;日间 RSD分别为 4 .4 8%、2 .6 4 %。
A method for the determination of silybin in human plasma by RP-HPLC was established. Plasma protein was precipitated with acetonitrile, centrifuged, and the supernatant 10 μl injection analysis. Chromatographic conditions: C18 column (10 μm) 220 mm × 4.6 mm; mobile phase: V (methanol): V (water): V (0.5 mol · L -1 potassium dihydrogen phosphate) 10: 1 (adjusted to pH 2.0 with 0. 2 mol · L-1 phosphate); flow rate 1.0 ml · min-1; detection wavelength 2 90 nm. Results The average recoveries of silybin A and B were 98.96% and 97.93%, respectively. The intraday RSD were 3.08% and 2.15% respectively. The daytime RSD were 4.48% and 2.64% respectively.
