对脂质体介导的体外肺癌细胞DAN转效率的差异性进行研究。方法 :提取质粒 pCEAcd/tk ,脂质体包裹后转染 6种肺癌细胞株 ,G418筛选 ,计数细胞阳性克隆数。结果 :6种肺癌细胞株中 ,导入了pCEAcdt/tk并获得稳定表达的细胞阳性克隆数差异较大 ;脂质体ESCORT -DNA混合物与细胞孵育时间不同对转染效率也有一定的影响。结论 :脂质体介导的方法把融合自杀基因表达体系导入到肺癌细胞的效率存在细胞差异性
The liposome-mediated in vitro transfection efficiency of lung cancer cells was investigated. METHODS: Plasmid pCEAcd/tk was extracted and transfected into six lung cancer cell lines after liposome encapsulation. Screening was performed with G418 and the number of positive clones was counted. RESULTS : Among the six lung cancer cell lines, the number of positive clones with stable expression was significantly different when pCEAcdt/tk was introduced. The difference in liposome ESCORT-DNA mixture and cell incubation time also had a certain effect on transfection efficiency. Conclusion : The liposome-mediated approach to the fusion of suicide gene expression system to the efficiency of lung cancer cells exists cell differences
