目的探讨应用一次性密封栓封堵骨孔制作兔骨肿瘤模型对肿瘤细胞外渗的影响。方法取传代VX2肿瘤荷瘤兔1只,取出肿瘤并制成新鲜细胞悬液,分别植入20只成年新西兰兔双侧胫骨内。将20只兔左侧胫骨设为实验组,应用一次性密封栓封堵骨孔;右侧胫骨设为对照组,应用骨蜡封堵骨孔。模型制备14d后,2组取骨孔周围软组织行组织病理学检查,观察有无肿瘤细胞外渗情况,比较2组肿瘤细胞经骨孔转移率。结果实验组肿瘤细胞经骨孔转移率(15.0%)明显低于对照组(45.0%),差异有统计学意义(P<0.05)。结论应用一次性密封栓封堵骨穿孔制作兔骨肿瘤模型可明显改善肿瘤细胞外渗,是一种较理想的造模方式。
Objective To investigate the effect of using a one-off seal plug to make bone tumor in rabbit bone tumor model to extravasate tumor cells. Methods Tumor-bearing VX2 tumor-bearing rabbits were taken from one of the rabbits. Tumors were removed and fresh cell suspensions were prepared and implanted into the tibia of 20 adult New Zealand rabbits respectively. The left tibia of 20 rabbits as experimental group, a one-time sealing plug to seal the bone hole; the right tibia as a control group, the application of bone wax bone closure. After the model was prepared for 14 days, the soft tissue around the bone hole was removed for histopathological examination in two groups to observe the presence or absence of extravasation of tumor cells. The metastasis rate of tumor cells in the two groups was compared. Results The metastasis rate of tumor cells in the experimental group (15.0%) was significantly lower than that in the control group (45.0%), the difference was statistically significant (P <0.05). Conclusion The application of a one-time sealing plug to seal the perforation of rabbit bone tumor model can significantly improve the extravasation of tumor cells, which is an ideal mode of modeling.
