目的探讨CD14基因启动子区-159T/C、-1145G/A、-1359G/T三个SNP位点的多态性和新疆维吾尔族人群支气管哮喘(简称哮喘)的关系。方法 126例明确诊断的哮喘患者为哮喘组,按1:1配对法收集同性别、族别、居住地、年龄(相差5岁以内)的健康体检者为对照组。采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测各组-159T/C、-1145G/A、-1359G/T位点的多态性。结果 CD14基因中-159T/C、-1145G/A、-1359G/T三个SNP位点基因型频率和等位基因频率在哮喘组和对照组的分布差异无显著性(P>0.05)。-159T/C和-1145G/A位点等位基因频率在维吾尔族、汉族和高加索人群总体分布上差异有统计学意义(χ2=11.295,P=0.004和χ2=13.029,P=0.001)。结论 CD14基因-159T/C、-1145G/A、-1359G/T三个SNP位点基因型频率和等位基因频率分布与维吾尔族人群哮喘无直接关联。
Objective To investigate the polymorphisms of -159T / C, -1145G / A and -1359G / T in CD14 gene promoter region and bronchial asthma (asthma) in Xinjiang Uygur population. Methods A total of 126 asthmatic patients were diagnosed as asthma group. According to the 1: 1 pairing method, healthy subjects with the same sex, ethnicity, residence, age (within 5 years of age) were collected as the control group. The polymorphisms of -159T / C, -1145G / A and -1359G / T in each group were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results There was no significant difference in the frequencies of genotypes and alleles of -159T / C, -1145G / A and -1359G / T among CD14 genes in asthma group and control group (P> 0.05). The frequencies of alleles at -159T / C and -1145G / A loci were statistically different between Uygur, Han and Caucasians (χ2 = 11.295, P = 0.004 and χ2 = 13.029, P = 0.001). Conclusion There is no direct correlation between genotype frequency and allele frequency distribution of CD14 gene -159T / C, -1145G / A and -1359G / T SNPs and Uygur population asthma.
