川芎嗪和参麦注射液对老龄大鼠脑缺血再灌注多器官损伤的作用

目的 :探讨老龄大鼠脑缺血再灌注所致多器官损伤机制 ,观察川芎嗪和参麦注射液及其配伍对多器官损伤的保护作用。方法 :老龄 (2 0月龄以上 )大鼠分为模型组、对照组、尼莫通组、川芎嗪组、参麦组和川芎参麦组 (川参组 ) ,检测各组药物对老龄大鼠全脑缺血 30分钟及再灌注 6 0分钟后多巴胺 (DA)、去甲肾上腺素(NE)、皮质醇 (Co)、一氧化氮 (NO)和肿瘤坏死因子 (TNF)水平 ,同时观察脑、心、肝、肺、肾、胃、小肠病理改变。结果 :模型组大鼠脑组织均出现病理改变 ,交感肾上腺系统兴奋增强 ,脑组织中 TNF含量增高 ,血清中 NO含量降低。用药各组老龄大鼠脑组织损伤均有明显减轻 ,下丘脑 NE和脑组织 NO含量增高及下丘脑 DA水平降低 ,川芎嗪组、参麦组及川参组下丘脑 NE含量和 DA水平接近对照组 ,参麦组血中 Co水平和川参组脑组织TNF含量降低 ,川芎嗪组血清中 NO水平增高。模型组大鼠多个器官较对照组出现病理改变 ,交感肾上腺系统兴奋增强 ,各组药物对器官损伤均有不同程度的保护作用和对交感肾上腺系统有明显的调节效果。结论 :老龄大鼠脑缺血再灌注损伤与交感肾上腺系统兴奋增强和 NO含量降低及 TNF增高有关 ,川芎嗪和参麦注射液及其配伍通过调节交感肾上腺系统和提高 NO水平以防护老龄大鼠脑缺血再灌注脑组? Objective : To explore the mechanism of multiple organ damage induced by cerebral ischemia-reperfusion in aged rats and to observe the protective effects of tetramethylpyrazine and Shenmai injection on their compatibility with multiple organs. METHODS: Older age (20 months or older) rats were divided into model group, control group, Nimotop group, Ligustrazine group, Shenmai group and Chuanxiong Shenmai group (Chuanshen group). The levels of dopamine (DA), norepinephrine (NE), cortisol (Co), nitric oxide (NO), and tumor necrosis factor (TNF) were measured after 30 minutes of global cerebral ischemia in rats and after 60 minutes of reperfusion. Brain, heart, liver, lung, kidney, stomach, small intestine pathological changes. RESULTS: The brain tissue of rats in the model group showed pathological changes, the sympathetic adrenal system increased in excitotoxicity, the TNF content in brain tissue increased, and the serum NO content decreased. The injury of brain tissue in all groups of aged rats was significantly reduced, and the content of NO in hypothalamus NE and brain tissue was increased and the level of DA in hypothalamus was decreased. The NE content and DA level in hypothalamus of tetramethylpyrazine group, Shenmai group and Chuanqi group were similar to the control group. In the Shenmai group, the level of Co in the blood and the content of TNF in the brain tissue of the Chuanshen group decreased, and the level of NO in the serum of the Ligustrazine group increased. Rats in the model group had pathological changes in multiple organs compared with the control group, and the sympathetic adrenal system was more excited. Each group of drugs had different degrees of protective effects on organ damage and had significant regulatory effects on the sympathetic adrenal system. Conclusion: The cerebral ischemia-reperfusion injury in aged rats is related to the enhancement of sympathetic adrenal system excitement and the decrease of NO content and TNF elevation. Ligustrazine and Shenmai injection and their compatibility can regulate the sympathetic adrenal system and increase the level of NO to protect the aged rats.
