目的:观察桂枝茯苓汤对乳腺癌细胞株MCF-7的凋亡相关蛋白和细胞周期的影响。方法:将浓度为300μg/m L、900μg/m L的桂枝茯苓汤分别作用MCF-7细胞24 h、48 h后,采用四甲基偶氮唑盐法(MTT)法检测细胞抑制率,流式细胞仪方法检测MCF-7细胞周期各时向DNA含量的变化,细胞免疫化学方法检测线粒体凋亡通路相关蛋白Bcl-2、Bax、Caspase-3/9和Bid的表达情况。结果:与空白对照组比较,桂枝茯苓汤各浓度组细胞抑制率明显升高(P<0.05),其中G0/G1期细胞DNA含量明显升高(P<0.05),桂枝茯苓汤可以降低Bcl-2蛋白的表达,同时可以促进Bax、Caspase-3/9和Bid蛋白的表达。结论:桂枝茯苓汤能体外诱导乳腺癌细胞凋亡,其机制可能与线粒体凋亡通路有关。
Objective: To observe the effects of Guizhi Fuling Decoction on apoptosis-related proteins and cell cycle of breast cancer cell line MCF-7. Methods: MCF-7 cells were treated with Guizhi Fuling Decoction 300μg / m L and 900μg / m L for 24 h, respectively. Forty-eight hours later, MTT assay was used to determine the cell inhibitory rate, The changes of DNA content in MCF-7 cell cycle were detected by flow cytometry. The expressions of Bcl-2, Bax, Caspase-3/9 and Bid in mitochondrial apoptosis pathway were detected by immunocytochemistry. Results: Compared with the blank control group, the cell inhibition rate of Guizhi Fuling Decoction group was significantly increased (P <0.05), and the DNA content of G0 / G1 phase cells was significantly increased (P <0.05), Guizhi Fuling Decoction could be reduced Bcl-2 protein expression, at the same time can promote Bax, Caspase-3/9 and Bid protein expression. Conclusion: Guizhi Fuling Decoction can induce breast cancer cell apoptosis in vitro, which may be related to the mitochondrial apoptosis pathway.
