[目的]分析抗肺癌单抗12H6的靶抗原在肺癌细胞系和肺癌组织中的表达,研究单抗12H6体内外生物学功能。[方法]细胞免疫荧光、流式细胞荧光法检测单抗12H6靶抗原在3种肺癌细胞系中的表达及其定位,免疫组化法检测其在人肺癌组织中表达及特异性;Transwell法和CCK-8法分别检测12H6体外对肺癌细胞系侵袭和增殖功能的影响。共接种模型裸鼠体内治疗实验研究12H6对移植瘤生长的抑制作用。[结果]单抗12H6的靶抗原在人肺腺癌细胞(A549、GLC-82)、人肺鳞癌细胞(GLC-P)的胞内及胞膜均有表达,在3种细胞系胞膜的表达比例分别为31.2%、15.4%和16.7%。单抗12H6在人肺癌组织中的表达比例为80.6%,较配对癌旁组织显著上调。单抗12H6在体外能明显抑制肺癌细胞侵袭和增殖;体内能明显抑制移植瘤生长,抑制率为40%。[结论]单抗12H6具有优良的体内外抑瘤功能,可能为肺癌靶向治疗提供潜在的靶向治疗药物。
[Objective] To analyze the expression of anti-lung cancer monoclonal antibody 12H6 in lung cancer cell lines and lung cancer tissues and to study the biological functions of monoclonal antibody 12H6 in vitro and in vivo. [Methods] The expression and localization of 12H6 mAb in three kinds of lung cancer cell lines were detected by immunofluorescence and flow cytometry. The expression and specificity of 12H6 were detected by immunohistochemistry in human lung cancer tissues. Transwell method and CCK-8 assay was used to detect the effect of 12H6 on invasion and proliferation of lung cancer cell lines in vitro. Co-vaccination model nude mice in vivo treatment of experimental study on the inhibitory effect of 12H6 on the growth of transplanted tumors. [Results] The target antigen of mAb 12H6 was expressed both intracellularly and membranely in human lung adenocarcinoma cells (A549, GLC-82) and human lung squamous cell carcinoma cells (GLC-P) The expression ratios were 31.2%, 15.4% and 16.7% respectively. The ratio of monoclonal antibody 12H6 expression in human lung cancer was 80.6%, which was significantly up-regulated compared with matched paracancerous tissues. Monoclonal antibody 12H6 could obviously inhibit the invasion and proliferation of lung cancer cells in vitro; the growth of the transplanted tumor was inhibited obviously in vivo with the inhibition rate of 40%. [Conclusion] McAb 12H6 has excellent antitumor activity in vivo and in vitro, which may provide potential therapeutic target for lung cancer targeted therapy.
