内膜增生是移植静脉失败的主要原因.发病机理尚不清楚。本研究取Wistar大鼠80只建立自体颈静脉移植于肾下腹主动脉动物模型.通过电镜和组织切片原位杂交分期于术后2h、24h及术后1周观察移植静脉超微结构和血小板源生长因子(PDGF)及受体基因表达动态变化。结果显示:(1)术后2h.内膜破裂翻卷,SMC线粒体轻度肿胀;PDGF-A及PDGFR-βmRNA表达增加.但PDGF-BmRNA表达量较大;(2)术后24h.大部分内皮细胞脱落,内膜被覆以丝状血栓;PDGF-A与PDGFR-β较术后2h表达明显增加;(3)术后1周,内皮开始重新再生;PDGF-AmRNA表达开始减少.PDGFR-β持续表达。结果表明PDGF在自体静脉移植中与SMC增殖及内膜增生有关。
Intimal hyperplasia is the main reason for graft failure. The pathogenesis is not clear. In this study, 80 Wistar rats were established autologous jugular vein graft in the subretinal aorta animal model. The changes of ultrastructure, expression of platelet-derived growth factor (PDGF) and receptor gene were observed by electron microscopy and in situ hybridization in situ at 2h, 24h and 1h after operation. The results showed: (1) 2h after surgery. Endometrial rupture roll, SMC mitochondria mild swelling; PDGF-A and PDGFR-βmRNA expression increased. But the expression of PDGF-B mRNA is larger; (2) 24h after operation. Most of the endothelial cells were shedding and the endothelium was covered with filamentous thrombus. The expression of PDGF-A and PDGFR-β was significantly increased at 2h after operation. (3) The endothelial began to regenerate one week after operation, and the expression of PDGF-AmRNA began to decrease. PDGFR-β continued to be expressed. The results indicate that PDGF is involved in SMC proliferation and intimal hyperplasia in autologous vein grafts.
