目的:探讨抑癌基因p53和癌基因mdm2在肺腺癌细胞系GLC82中的相互关系和作用.方法:采用脂质体Lipofetin介导的DNA转染法,用含有野生型p53基因和mdm2基因的PDORneo逆转录病毒载体分别或共转染GLC-82细胞.结果:转染野生型p53基因可阻止GLC82细胞生长;抑制GLC82细胞DNA合成;软琼脂培养集落形成率减少及裸小鼠成瘤性减慢,而mdm2基因的转染可拮抗野生型p53的功能.结论:将mdm2表达载体转染含有野生型p53基因的GLC82细胞后,mdm2可部分地解除野生型p53对GLC82细胞生长的抑制作用.
Objective: To investigate the relationship between tumor suppressor gene p53 and oncogene mdm2 in lung adenocarcinoma cell line GLC82. METHODS: Lipofetin-mediated DNA transfection was used to transfect GLC-82 cells with PDORneo retroviral vectors containing wild-type p53 and mdm2 genes. RESULTS: Transfection of wild-type p53 gene prevented the growth of GLC-82 cells; inhibited DNA synthesis in GLC-82 cells; soft agar culture reduced the rate of colony formation and tumorigenesis in nude mice; transfection of mdm2 gene could antagonize the wild P53 function. Conclusion: After mdm2 expression vector was transfected into GLC82 cells containing wild-type p53 gene, mdm2 could partially relieve the inhibitory effect of wild-type p53 on the growth of GLC82 cells.
