目的研究发酵乳酸杆菌(Lactobac ilus ferm entum,Lb-f)对人结肠癌(HT-29)裸鼠异种移植瘤生长抑制作用及抗肿瘤机制。方法用HT-29裸鼠异种移植瘤模型治疗实验,观察Lb-f抗人结肠癌作用;碳粒廓清法、足跖厚度增加法和小鼠抗绵羊红细胞凝集素抗体滴度测定,观察Lb-f对昆明种(KM)小鼠细胞免疫及体液免疫功能的调节作用。结果Lb-f对HT-29裸鼠皮下移植瘤的生长抑制率分别为63%、79%、83%,瘤重抑制率分别为44%、49%、71%;Lb-f作用KM小鼠,校正碳粒廓清指数α值分别为:2.3928±0.3941,3.4092±0.1711,4.5036±0.215,1.9350±0.8839,P<0.05;足跖肿差分别为:(1.01±0.07)mm,(1.37±0.09)mm,(2.01±0.14)mm,(0.45±0.05)mm,P<0.001;小鼠抗绵羊红细胞抗体滴度增加,分别为:336±176,544±154,832±309,100±43,P<0.01。结论Lb-f对HT-29细胞裸鼠异种移植瘤生长抑制作用与提高机体免疫功能相关。
Objective To investigate the inhibitory effect of Lactobacillus ferus (Lb-f) on xenograft tumor in human colon cancer (HT-29) nude mice and its anti-tumor mechanism. Methods The model of HT-29 xenograft in nude mice was used to treat Lb-f anti-human colon cancer cells. The carbon clearance, the increase of plantar thickness and the antibody titer of mouse anti-sheep hemagglutinin were observed. The effects of Lb- f on the regulation of cellular immunity and humoral immune function in Kunming (KM) mice. Results The inhibitory rates of Lb-f on the growth of HT-29 xenografts in nude mice were 63%, 79% and 83%, respectively. The rates of tumor weight inhibition were 44%, 49% and 71% (1.01 ± 0.07) mm, (1.37 ± 0.09) mm, respectively. The corrected α value of carbon particle clearance index was 2.3928 ± 0.3941,3.4092 ± 0.1711,4.5036 ± 0.215,1.9350 ± 0.8839, P < (2.01 ± 0.14) mm, (0.45 ± 0.05) mm, P <0.001. The titer of mouse anti-sheep erythrocyte antibody increased: 336 ± 176,544 ± 154,832 ± 309,100 ± 43, P <0.01. Conclusions The inhibitory effect of Lb-f on the growth of xenografts in nude mice of HT-29 cells is related to the enhancement of immune function.
