目的 研究转化生长因子 β(transforminggrowthfactor β ,TGF β)对人牙周膜成纤维细胞(humanperiodontalligamentfibroblasts,HPDLFs)的生物学作用。方法 原代培养HPDLFs,并观察TGF β对HPDLFs的增殖、碱性磷酸酶 (alkalinephosphatase ,ALP)活性、骨钙素 (osteocalcin ,OC)合成及矿化结节形成的作用。结果 0 0 0 5 0~ 0 10 0 0mg/LTGF β可明显刺激HPDLFs的增殖 ,0 0 0 5 0mg/LTGF β可显著提高HPDLFs的ALP活性 ,0 0 0 0 5~ 0 10 0 0mg/LTGF β对HPDLFs合成OC和矿化结节的形成无明显影响。结论 TGF β对HPDLFs的作用呈剂量依赖性。TGF β能够刺激HPDLFs表达某些成骨细胞表型 ,但不能促进成骨表型的成熟。
Objective To study the biological effects of transforming growth factor β (TGF β) on human periodontal fibroblasts (HPDLFs). Methods HPDLFs were cultured in primary culture. The effects of TGFβ on the proliferation of HPDLFs, alkaline phosphatase (ALP) activity, osteocalcin (OC) synthesis and mineralized nodule formation were observed. Results 0 0 0 0 0 0 0 0 0 0 0 0 0 0 mg / L TGFβ significantly stimulated the proliferation of HPDLFs. ALD activity of HPDLFs was significantly increased by 0 0 0 0 0 0 0 0 mg / L TGFβ, 0 0 0 0 0 0 0 0 0 0 0 mg / There was no significant effect on the formation of OC and mineralized nodules in HPDLFs. Conclusion The effect of TGF β on HPDLFs is dose-dependent. TGFβ stimulated the expression of certain osteoblastic phenotypes in HPDLFs but did not promote the maturation of osteogenic phenotypes.
